Universal Lateral Flow Assay Kit for DNA and RNA Detection
Attogene’s Universal Lateral Flow Assay Kit is a ready-to-use dipstick kit for building a rapid lateral flow readout for labeled nucleic acids, including amplification products from PCR, LAMP, RPA, and CRISPR (Cas12a) workflows. The strips arrive pre-striped, so you can test your labeled product on day one. You don’t need to stripe capture antibodies or buy striping equipment. It’s a fast, low-cost way to prove feasibility before you invest in custom lateral flow development.
How it works
Each dipstick carries 40 nm colloidal gold conjugated to streptavidin, which binds any biotin-labeled molecule. Your product must carry two labels: biotin (binds the gold) and one capture label (FITC/FAM, DIG, or DNP, which binds a test line). Dual-labeled product is captured at its test line and forms a red-purple line. Unbound gold continues to the biotin-BSA control line.
Choose your format
| Option | Catalog # | Test lines | Targets per strip |
|---|---|---|---|
| 2 Line | AU2034-01 | anti-FITC/FAM | 1 |
| 3 Line | AU2034-02 | anti-FITC/FAM, anti-DIG | 2 |
| 4 Line | AU2034-03 | anti-FITC/FAM, anti-DIG, anti-DNP | 3 |
All formats include a biotin control line, with lines spaced 4.5 mm apart. Need a different configuration or a cassette format? Contact sales@attogene.com.
Kit components (50 tests)
- 50 lateral flow dipsticks (4.5 mm × 60 mm), in a desiccated canister
- 12 mL RNase-free nucleic acid running buffer
- 20 µL positive control nucleic acid per capture label (biotin + FAM; plus biotin + DIG and biotin + DNP in 3- and 4-line kits)
- 96-well plate and manual
Protocol at a glance
- Add 150 µL running buffer to a well.
- Add 1–5 µL of your labeled product (keep it under 15 µL) and mix.
- Insert a dipstick, dip end down.
- Read at 15 minutes by eye, with a photo, or with a lateral flow reader.
Run a positive control (5 µL control oligo) and a buffer-only negative control with every run.
Reading results
- Test line present: labeled target detected. Line intensity increases with the amount of product.
- Control line: it fades as test lines strengthen. That’s normal, because test lines capture the gold first.
- Both lines faint at high input: this is the hook effect. Dilute your sample and re-run.
- No lines: the strip didn’t run. Repeat with a new strip.
Primer design tips
- PCR: put biotin on the 5′ end of one primer and the capture label (FAM, DIG, or DNP) on the 5′ end of the opposite primer.
- LAMP / RPA: place both labels on primers or a probe that end up in the same double-stranded product.
- Multiplex: biotin goes on one primer of each target, and each target gets its own capture label.
- Always include a no-template control. Primer-dimers carrying both labels can produce a test line.
Features and benefits
- Pre-striped strips, so no antibody striping or equipment is needed
- Multiplex up to 3 targets on one strip
- Results in 15 minutes, read visually or with a reader
- Strips and buffer store at room temperature (controls at 2–8 °C)
- Add-ons available: extra running buffer, control nucleic acids, and cassetted strips (CO2022-02)
Frequently asked questions
What labels do I need?
Biotin plus one of FITC/FAM, DIG, or DNP on the same product.
Can I use it with LAMP or RPA?
Yes. It’s designed for amplified DNA/RNA from PCR, LAMP, and RPA.
Can it detect more than one target?
Yes. The 3-line kit detects 2 targets and the 4-line kit detects 3.
Is it quantitative?
It’s qualitative or semi-quantitative. Line intensity can be measured with a lateral flow reader.
Can Attogene build a custom assay from my results?
Yes. See our Lateral Flow Development Services.
