MTBC species share >99.9% genomic identity, making species differentiation challenging. A key genetic target for molecular detection is the IS6110 insertion sequence, a multicopy, mobile element present in most MTBC strains but largely absent in non-tuberculous mycobacteria. IS6110s high copy number and species specificity make it a sensitive and reliable marker for PCR-based diagnosis of TB across diverse clinical specimens.
